Explanation
This comic takes the classic Peter Pan scene literally. In the first panel, Peter Pan cheerfully tells the children "Second star to the right and straight on till morning!" -- his famous directions to Neverland from J.M. Barrie'''s story. The children look excited and ready for adventure.
In the second panel, we see the realistic consequence of following these directions: the children are floating in the cold void of outer space, panicked and clearly in danger. Stars are, of course, unfathomably distant celestial objects, not waypoints you can casually fly toward. Taking Peter Pan'''s whimsical directions at face value would mean flying into the deadly emptiness of space rather than arriving at a magical island. The humor comes from applying real-world physics and astronomy to a beloved children'''s fairy tale.
The votey panel shows someone (likely Zach Weinersmith) writing with the caption "Peter Pan is unrealistic!" -- a self-aware acknowledgment that the comic'''s entire premise is pointing out the obvious absurdity in a fantasy story, played for laughs.
