Explanation
This comic takes the concept of a "dad joke" and turns it into something much darker and more absurd.
In the first panel, a man solemnly tells his child that he has something to confess. He got his DNA tested and discovered that he is "not biologically yours." This is already a funny inversion of the typical revelation -- normally a parent would tell a child "you're not biologically mine," but here the father reverses the possessive, as if the child owns the parent rather than the other way around.
The father then launches into the ultimate dad joke: "Hi, Not Biologically Yours, I'm Da--" -- attempting the classic dad joke format of deliberately misinterpreting "I'm [adjective]" as a name introduction ("Hi [adjective], I'm Dad"). But he can't even finish the word "Dad" because the premise of his own joke undermines his claim to the title.
The final panel shows the father literally running away into the darkness while the child watches, arms outstretched. He set up an elaborate scenario purely for the sake of a dad joke and then fled when it collapsed under its own logic. The humor comes from the absurd commitment to the bit -- engineering a dramatic confession scene just to set up a pun, and the dark visual of a father abandoning his child as the punchline.
