Explanation
The Joke
A religious figure (resembling a priest or pastor) declares that "God did not intend for man to lay as one with other men." When challenged with "How do you think you know?" he responds that if God had intended for men to have sex, he would have made them "more flexible and also in order to mumble" -- trailing off as he inadvertently starts describing how male-male sex could work, implying he has thought about this in considerable detail. His interlocutor observes that "sex with two dongs at once would be a significant improvement over the status quo," and the priest, now unable to contain himself, says "I can't tell which of us was supposed to be the kicker" -- meaning he's lost track of who was supposed to be arguing which side. The final panel shows them apparently making out, with one saying "making out with other men is fine. One of us was making that point."
The comic satirizes anti-gay religious arguments by showing that the person making the argument against homosexuality is actually more enthusiastic about the concept than the person challenging him, which is a common satirical trope about repressed desire among vocal homophobes.
The Humor
The humor comes from the rapid unraveling of the religious figure's anti-gay stance. What starts as a familiar homophobic sermon collapses the moment he actually thinks through the logistics, and his attempt to argue against gay sex inadvertently becomes an argument for it. The joke escalates beautifully: the priest goes from condemning homosexuality, to accidentally praising it, to being unable to remember which side he was on, to actively participating in it -- all within four panels. The phrase "one of us was making that point" in the final panel is a perfect absurdist capper, as they have abandoned the debate entirely.
